Job Description This project aims to study ciliopathies such as Primary Ciliary Dyskinesia (PCD) and Polycystic Kidney Disease (PKD). By utilising human stem cell-derived models, the research seeks to further characterise these genetic disorders and evaluate potential treatments via gene therapy approaches. • Maintenance of human pluripotent stem cell lines and multi-step directed differentiation to generate lung and kidney tissues, including the use of 3D organoids, air-liquid interface (ALI) cultures, and Flow Cytometry (FACS) for the characterisation and isolation of specific cell populations. • Proficiency in core molecular biology techniques including cloning and PCR-based analysis for genetic characterisation. • Production, titration, and application of viral vectors for gene editing and stable expression within differentiated cell models. • Characterisation of ciliary morphology and protein localisation using multi-channel immunofluorescence and high-resolution confocal microscopy. Job Requirement • Academic Background: PhD or Master’s degree in Biomedical Sciences, Molecular Biology, Bioengineering, or a related life sciences discipline. • Research Focus: Proven track record in stem cell biology or regenerative medicine is highly preferred. • Professional Experience:For PhD holders: 0–3 years of postdoctoral experience Technical Competencies • Cell Culture Mastery: Extensive experience in maintaining human pluripotent stem cells (hPSCs) and performing directed differentiation. • Tissue Engineering: Proficiency in 3D organoid culture and/or Air-Liquid Interface (ALI) models. • Gene Editing & Virology: Hands-on experience with viral vector production (Lentivirus/AAV) and molecular cloning techniques. • Analytical Techniques: Skilled in Flow Cytometry (FACS) for cell sorting and analysis.
